Evaluation of cardiovascular, respiratory, and gas exchange parameters in a rat model of acute respiratory distress syndrome

Background and Aim: Animal models are essential for investigating the pathophysiology of acute respiratory distress syndrome (ARDS) and evaluating potential therapeutic interventions. This study aimed to evaluate a modified method of inducing acute lung injury (ALI) using repeated saline lavage and to characterize the associated cardiovascular, respiratory, and gas exchange changes in rats.
Methods: Male rats were divided into sham and saline lavage-induced lung injury groups. Following tracheostomy and femoral artery cannulation, the animals were mechanically ventilated using a volume-controlled mode. Lung injury was induced by 10 consecutive saline lavage procedures, each using 3 mL of normal saline, performed over 1 hour, followed by a 1-hour recovery period. Mean arterial pressure (MAP), heart rate (HR), maximum inspiratory pressure (MIP), maximum expiratory pressure (MEP), and arterial blood gas parameters were assessed.
Results: Compared with the sham group, the saline lavage group showed significant decreases in MAP and HR and significant increases in MIP and MEP (p < 0.05). These changes remained relatively stable throughout the recovery period. In addition, arterial oxygen tension (PaO2) and the PaO2/FiO2 ratio decreased, whereas arterial carbon dioxide tension (PaCO2) increased in the saline lavage group (p < 0.05), demonstrating impaired pulmonary gas exchange characterized by hypoxemia and hypercapnia.
Conclusion: The modified repeated saline lavage protocol, consisting of 10 lavage procedures over 1 hour, produced sustained acute lung injury in rats. This model may be useful for investigating the pathophysiological mechanisms of ARDS and evaluating potential therapeutic interventions. |
