fa نگاهی دوباره به تاثیر ویروس هاری بر سیستم عصبی با تاکید بر نقش گلیکوپروتئین ویروسی A new look at the effect of rabies virus on the nervous system with emphasis on the role of viral glycoprotein علوم اعصاب Neuroscience مروری-با دعوت سردبیر Review Article -by invitation بیماری هاری توسط ویروس نوروتروپیک هاری از جنس لیساویروس و از خانواده رابدو ویریده ایجاد می‌شود. علیرغم وجود علائم بارز و مشخص رفتاری همچون اضطراب، رفتارهای خشونت‌آمیز، توهم، افسردگی و ترس از آب، هاری با ضایعات بافتی فاحشی در مغز همراه نیست. <a name="_Hlk88507269">ساختار ویروس هاری از پنج پروتئین با نامهای </a>N، L، P، M و G تشکیل شده است. گلیکوپروتئین G بخشی از خارهای سطح خارجی ویروس را ایجاد می‌کند که با اتصال و هم‌جوشی به غشای سلول موجب ورود ویروس به داخل سلول می‌شوند. بخشی از انتهای کربوکسیل گلیکوپروتئین G که PBM (PDZ binding motif)  خوانده می‌شود با فعال‌کردن آبشارهای سیگنالینگ داخلسلولی باعث تغییر عملکرد سلول میزبان و بیماریزایی ویروس هاری می‌گردد. گلیکوپروتئین ویروس هاری هر دو نوع شکل‌پذیری سیناپسی کوتاه‌مدت و بلندمدت را تسهیل می‌کند، عملکرد حافظه فضایی وابسته به هیپوکمپ را افزایش می‌دهد و رفتار اضطرابی در موشهای آلوده را موجب می‌شود. در این مقاله، نحوه تاثیر ویروس هاری بر عملکرد سیناپسی، استفاده از آن جهت ردیابی شبکه‌های عصبی و دارورسانی به سیستم عصبی مرکزی را مرور خواهیم کرد Rabies is caused by the neurotropic rabies virus from the genus Lyssavirus belonging to the Rhabdoviridae family. Despite extensive neurological symptoms including anxiety, rage, hallucinations, depression and hydrophobia, mild pathological alterations are usually seen in the rabies-infected brain. Rabies virus encodes five proteins named N, L, P, M and G. Glycoprotein G forms spikes on the external surface and enables the virus to enter the host cells by connecting and fusing with the cell membrane. The C-terminus amino acid residue of glycoprotein G, called PBM (PDZ binding motif), determines the viral pathogenesity and may manipulate different signaling pathways in the infected neurons. Rabies virus glycoprotein G has been shown to facilitate both short-term and long-term synaptic plasticities, enhances hippocampal-dependent spatial memory performance, and triggers anxious behavior in the infected rats.
